Meaning

An agreement can be expressed in various ways, periodt is one such way. The phrase simply means to agree enthusiastically. While a person can simply say, yes, or I agree, the slang term periodt is intended to convey a heavier form of agreement than a simple yes.

The phrase is also meant to end further conversation on a specific topic. It is usually stated with vigor and a small amount of aggression to get one's point across.

When you want to end a conversation, even if you are not in agreement, using the term is also appropriate. It is also a way to casually emphasize a point or a comment during a heated conversation between two or more people. In this instance, it does not always mean agreement, more so that you feel the point being made is worth listening to. More information can be found in the video (here).
